Van Dijk, who picked up the knock during Sunday’s 3-0 Premier League win over Leicester City, faces a late test according to manager Claude Puel.
Having already seen captain Jose Fonte leave the club in a £8million transfer to West Ham, Van Dijk’s absence could leave Paul desperately short in defense as the Saints try to defend their one goal advantage.
‘Van Dijk, he is having a scan and I am waiting for the results at the moment,’ Paul told the Southampton’s official website.
‘It’s not serious, but I don’t know if he can participate in this game. We will see in the next days before the game,’ he added.
In some slightly more encouraging news for Southampton fans, Steven Davis, who hasn’t played since picking up an injury against Liverpool in the first leg, has stepped up his recovery and could feature at Anfield on Wednesday night.
